SNX27 and the retromer complex
​

SNX27 is a PDZ-domain member of the sorting nexin family of endosomal trafficking components, and a member of the retromer complex.

Our previous studies indicate that SNX27 can enhance endosome to cell surface recycling of SORLA, and inhibit gamma-secretase dependent APP cleavage, both of which reduce Abeta generation.

​We are in the process of characterizing other SNX27 targets that may be relevant to AD.

Exploring neuroprotective roles for SORLA in neurons and microglia

SORLA is a transmembrane trafficking component which can associate with the SNX27/retromer complex.

SORLA polymorphisms have also been associated with increased AD risk. We are exploring why SORLA dysregulation might affect AD onset in neurons and microglia.

The microglial TREM2 immune receptor and AD

TREM2 is a microglial immune receptor we have shown to bind and mediate microglial activation in response to Abeta.

Mutations in TREM2 (R47H) have been associated with significantly increased risk of AD onset. We are currently investigating potential mechanisms underlying TREM R47H and TREM2-associated AD pathogenesis.

We are also interested in proteostasis, protein misfolding and neurodegeneration. We are currently investigating how UPR components potentially relate to AD, as well as potential relationships between the ERAD complex and the newly-described ERAD component membralin in neurodegeneration. We are also exploring potential roles for E3 ubiquitin ligases such as Siah1 in neurodegeneration.
